N-acetyl D-glucosamine (NAG) is an amino sugar and acetylated derivative of glucosamine that occurs naturally in human skin as a key precursor in the biosynthesis of glycosaminoglycans (GAGs), including hyaluronic acid. In topical skincare formulations, NAG is primarily valued for its skin-brightening, barrier-supporting, and anti-aging properties. Mechanistically, NAG has been shown to modulate melanogenesis by inhibiting tyrosinase glycosylation and reducing melanin transfer, leading to gradual improvement in uneven skin tone and hyperpigmentation. In parallel, its role as a substrate in hyaluronic acid synthesis supports epidermal hydration and structural integrity, contributing to smoother texture and enhanced skin resilience.

From a formulation perspective, N-acetyl D-glucosamine is water-soluble, chemically stable across a broad pH range, and highly compatible with other cosmetic actives, including niacinamide. Synergistic use with niacinamide has been extensively documented, demonstrating amplified improvements in pigmentation, fine lines, and barrier function compared to either ingredient alone. In hair and scalp care, NAG is less commonly used but may contribute indirectly to scalp barrier maintenance and hydration, particularly in leave-on scalp treatments formulated for sensitive or compromised skin. Its non-exfoliating mode of action and favorable tolerability profile make it suitable for long-term, daily-use formulations.
